GHRP-6 is a synthetic hexapeptide and one of the earliest discovered growth hormone secretagogues. It stimulates GH release from the pituitary by activating the ghrelin receptor (GHS-R1a). GHRP-6 is notable for causing a strong increase in appetite (via ghrelin pathway activation) and has been studied for GH deficiency diagnostics, wound healing, and cardioprotection.

GHRP-6 works like pressing the gas pedal on your growth hormone engine while also opening the fuel intake valve — it boosts GH output and simultaneously increases your appetite so your body has the raw materials to build and repair.

Mechanism · technical
GHRP-6 binds to the ghrelin/GHS-R1a receptor on pituitary somatotrophs, triggering intracellular calcium release and subsequent growth hormone secretion. It simultaneously suppresses somatostatin tone, amplifying GH pulse magnitude. Its strong activation of the ghrelin pathway also stimulates appetite through hypothalamic feeding centers and promotes gastric motility. GHRP-6 has demonstrated cardioprotective effects in ischemia-reperfusion models, likely through PI3K/Akt signaling.

Side effects, rare serious events, who shouldn't.

Reported side effects
The most notable side effect is a significant increase in appetite and hunger, typically occurring 20-30 minutes after administration. Other effects include water retention, transient increases in cortisol and prolactin (less than GHRP-2), potential blood sugar fluctuations, and injection site reactions. Some users report lightheadedness shortly after injection.
